Introduction
Fiber-optic evanescent wave biosensors FOB Series are constructed
based on the principle of evanescent wave while light propagates in an
optical fiber. When the target biomolecules in question are
specifically bound to the capture component immobilized on the fiber
core surface, the fluorophores labeled to the target biomolecules
become attached within the evanescent zone of the core surface at the
same time. Excitation of fluorophore in the evanescent zone occurs
when light is launched into the fiber. Fluorescent emission at a
longer wavelength is trapped in the fiber and is then coupled back
into the fiber. The fluorescent flux relates to the concentration of
target biomolecules.
This type of biosensors has several
advantages over other fluorescence-based sensors. The evanescent wave
penetrates essentially only a fraction of a wavelength into the sample
medium and thus excites fluorescence primarily from the fluorescently
labeled analyte complexes that have been bound to the surface through
specific antibody-antigen recognition reaction (i.e., immunoassay) and
hybridization of nucleic acid. Not only does this reduce the
fluorescence from the sample or unbound fluorescent reagents, but the
signal obtained is directly related to the binding kinetics of the
detection interaction, i.e., a rapid real-time response can be
observed without complicated washing or separation steps, and field
detection of a variety of analytes can be performed.
